It had originally been suggested that the first leg of the tie would be played on Wednesday, with the return on Sunday, but both matches will be played 24 hours earlier.
Stevie Crawford's side travel to already relegated Alloa Athletic tonight in the final match of the regular Championship season having already assured themselves of at least a fourth place finish.
The fourth placed side will face the team in third in the quarter final, but the identity of Dunfermline's opponents - and the venue for each leg - is still to be decided.
The Pars are three points behind third placed Dundee, and two goals worse off, but if they secure a margin of victory that overturns that, and the Dark Blues lose at Queen of the South, they'd finish third.
That would give them give them home advantage for the second leg, which will be played on Saturday May 8 at 3pm, with the fourth-top team at home on Tuesday at 7pm.
A point for Dundee will secure them third place but, if they win and Raith Rovers lose at home to champions Hearts, James McPake's side would finish second.
That would then mean a home game for Dunfermline with their Fife rivals on Tuesday, with the return in Kirkcaldy next Saturday.
The Pars have also announced that season ticket holders can watch a stream of the home leg for free on Pars TV.
#DAFC are pleased to announce that all of our season ticket holders will be provided with free access to our normal @ParsTV_Official streaming service for our home quarter final tie at East End Park.
