The tie will give the East Super League champions a shot at revenge after their opponents knocked them out of the First Round proper in 2015.
Kelty qualified for the senior tournament courtesy of their league success but will play in the East of Scotland League next season after deciding to the quit the junior set-up in favour of becoming an SFA licenced club.
Two years ago Kelty were given a bye in the first preliminary round before travelling to Civil Service Strollers, where they recorded a 4-0 win.
That set up a tie at Lothian Thistle Hutchison Vale in round one but a poor performance by Hearts saw them comprehensively knocked out by 3-0.
The game will be the first-ever senior Scottish Cup tie to be played in Kelty and player/boss Courts tweeted: "Great for the club and the local community to have a home tie to look forward to. Look forward to giving LTHV a proper Kelty welcome."
